目的: 观察安宫清开灵注射液对胶原酶诱导的自发性高血压大鼠( SHR )脑出血模型脑组织中多环节炎性细胞因子TNF-α、IL- 1β、ICAM-1和NF-κB 表达的影响, 探讨安宫清开灵注射液阻抑脑出血后炎症反应的作用环节以及作用效果。方法: 成年SHR 大鼠, 雌雄各半, 随机分成正常组、模型组、安宫清开灵低剂量组、安宫清开灵中剂量组、安宫清开灵高剂量组、阳性对照组(清开灵注射液)。基底核注射胶原酶法复制大鼠48h 脑出血模型。应用免疫组化法检测脑组织中TNF-α、IL-1β、ICAM-1和NF-κB的表达情况。结果: 脑出血48h, 大鼠脑组织TNF-α、IL-1β、ICAM-1以及NF-κB均有不同程度的表达。安宫清开灵中剂量组均能明显阻抑出血后48h脑组织TNF-α、IL-1β、ICAM-1和NF-κB表达的增加; 安宫清开灵低剂量组可以明显抑制ICAM-1和NF-κB的表达,而对TNF-α和IL-1β的作用不明显; 安宫清开灵高剂量组对TNF-α、IL-1β和ICAM-1表达的具有较强的抑制作用。结论: 安宫清开灵注射液对抑制脑出血后炎症因子和黏附分子的表达具有良好的作用, 从而能够减轻炎性损伤。其中以中剂量组作用效果尤为突出, 对脑出血后48h前炎症因子和黏附分子的表达都有显著的抑制作用, 可以在多环节干预炎症反应过程、阻抑脑出血后损伤级联反应从而发挥保护神经元的作用。

In this paper, authors studied the effects of An Gong Qing Kai Ling In ection on the expression o f TNF- α, IL-1β, ICAM-1 and NF-κB in the brain tissue of SHR having cerebral hemorrhage. They also discussed the links and effects of An Gong Qing Kai Ling Injection in inhibiting inflammatory reactions. In the study, the SHRs were randomly divided into six groups: normal group, model group, low-dose group, middle-dose group, high-dose group, and positive control group. Cerebral hemorrhage was induced using collagenase Ⅶ in SHR. Immunohistochemistry was employed to identify the expression of TNF-α, IL- 1β, ICAM-1 and NF-κB in the brain tissues of SHRs having cerebral hemorrhage. 48 hours after cerebral hemorrhage, TNF-α, IL- 1β, ICAM-1 and NF-κB produced an expression. The middle-dose group showed an obvious effect on inhibiting the increased expression of TNF-α, IL- 1β, ICAM-1 and NF-κB. The low-dose group can inhibit the increased expression of ICAM-1 and NF-κB, though having no apparent effect on TNF-αand IL- 1β. The high-dose group inhibited the increased expression of TNF-α, IL- 1β, and ICAM-1 in an apparentmanner. It is concluded that An Gong Qing Kai Ling Injection is of a noticeable effect on inhibit ing the expression of inflammatory factors and adhesionmolecules, which in turn alleviates the damage of inflammation. The middle-dose application can noticeably inhibit the expression of pro-inflammatory factors and adhesion molecule, 48 hours after cerebral hemorrhage, interfering inflammatory reactions in a number of links, and inhibiting the cascade reaction, a fter cerebral hemorrhage. It plays a role o f protecting the neurons.
