[摘要]
目的:利用基因芯片筛选双龙方主要成分(人参皂苷Rb1,丹参酚酸B)诱导大鼠骨髓间充质干细胞(MSCs)分化过程中的差异表达基因,并对差异表达基因进行聚类分析,研究双龙方主要有效成分对大鼠MSCs增殖及分化的影响。方法:用含5705条大鼠基因的博奥大鼠全基因组Oligo芯片,以不同给药情况、不同时间点的细胞总RNA制备的探针杂交芯片,用 LuxScan10KA双通道激光扫描仪(CapitalBio公司)进行扫描,筛选MSCs变化过程中的差异表达基因并进行生物信息分析,hierarchical聚类提取差异表达的特征基因。结果:在MSCs的分化过程中,筛选出128条(2.24%)差异表达基因,经分析发现它们与能量代谢,信号传导等多类基因密切相关,对样本进行聚类发现MSCs在20-30天之间发生了显著的改变。结论:基因芯片是研究中药作用机理的有效平台, 双龙方主要成分可诱导MSCs向心肌样细胞分化。

To investigate the effect of Shuanglong prescription components on MSCs by using gene chips to screening and cluster the differentially expressed MSCs genes induced by Shuanglong Prescription. METHODS: BOAO rat microarrays (include 5705 rat genes) were used to prepare gene chips hybrided by cell total RNA of different conditions. The chips were scanned with LuxScan 10KA dual channels laser scanner of CapitalBio Company to look for the information of differently expressed genes. RESULTS: 128 differently expressed genes(2.24%)were screened out from the 5705 target genes, which were found to be correlated with energy metabolism and signal transduction. Bioinformatics analysis suggests that MSCs changed remarkably in the period of 20 to 30 days. CONCLUSIONS: DNA microarray is an effective platform for the study and analysis of mechanisms of TCM. It can be concluded that Shuanglong Prescription components can induce MSCs to differentiate into cardiomyocytes.
