目的:从生物热活性变化的角度,考察不同干燥方式对板蓝根水提物质量的影响,为建立中药制剂工艺有无质的改变的早期快速检测方法体系提供依据。方法:板蓝根水提液分别采用冷冻干燥、减压干燥、常压干燥、喷雾干燥等4种不同方式进行干燥。通过测定各干燥样品作用于大肠杆菌的生物热活性图谱并拟合得到生物热动力参数,比较不同样品的抗菌生物活性的差异性。结果:不同干燥方式所得样品的生物热活性存在较大差异,喷雾干燥和冷冻干燥所得样品的作用明显强于减压干燥和常压干燥样品,提示干燥方式的改变对板蓝根水提物质量的影响不容忽视,当板蓝根制剂的干燥工艺从冷冻干燥、喷雾干燥转变成减压干燥或常压干燥时,须注意其生产工艺是否存在质的改变。结论:生物热活性检测具有普适性好、实时在线、灵敏准确、定性定量、经济高效等特点,可作为中药板蓝根水提物不同干燥工艺有无质的改变早期快速检测方法。

Authors investigated the impacts of different drying methods on the quality of Radix Isatidis water extracts in the context of biothermoactivity, in an attempt to provide evidences for establishing a system to detect the changed quality of TCM preparations in an early and fast manner. Radix Isatidis water extracts can be dried using a range of techniques, including frozen drying, vacuum drying, normal pressure drying, and spray drying. Biothermoactivities of the dried samples on E. coli were measured. Samples were compared for their antibacterial bioactivity with biothermodynamic parameters. Results show that Radix Isatidis water extracts may have quite different biothermoactivity when derived from different drying methods. For example, samples derived from spray drying and frozen drying have noticeably stronger biothermoactivity, compared with the samples treated otherwise. In this context, it is important to understand the impacts of different drying methods on the quality of Radix Isatidis water extracts. Quality can be deteriorated when drying methods are changed from frozen/spray drying to vacuum drying, and further to normal pressure drying. Biothermoactivity assay makes a method to evaluate the changed quality of TCM preparations in an early and fast manner, thanks to its fine adaptability, sensitivity, speed, and accuracy.
