目的:建立HPLC法测定参附强心丸中大黄素、大黄酚含量的方法。方法:色谱柱:Phenomenex GEMINI C18(5μm,4.6mm×250mm);流动相:甲醇-0.1%磷酸(75:25);检测波长:254nm;柱温:室温;流速:1.0mL·min-1。结果:大黄素线性范围为0.01098~0.549?滋g(r=0.9999),平均回收率为99.96%,RSD为0.56%,重复性RSD为0.70%(n=6),精密度RSD为0.15%(n=6),稳定性RSD为0.67%。大黄酚线性范围为0.03885~1.554?滋g(r = 0.9999),平均回收率为99.29%,RSD为0.32%,重复性RSD为0.59%(n=6),精密度RSD为0.63%(n=6),稳定性RSD为0.36%。结论:本文所得方法稳定、可靠,可作为该制剂的质量控制方法。

[Abstract]
This is an attempt to determine emodin and chrysophanol content in Shenfu Qiangxin Pill using HPLC. In the study, both Emodin and chrysophanol were assayed on a Phenomenex GEMINI C18(5μm, 4.6mm×250mm)column, with the mobile phase composed of methanol-0.1% phosphoric acid solution (75:25) at a flow rate of 1.0 mL·min-1. The determination wavelength was set at 254nm. Results indicate that the calibration curves were linear in a range from 0.01098ug to 0.549ug(r = 0.9999)for emodin, and from 0.03885ug to 1.554ug(r = 0.9999)for chrysophanol. The averaged recoveries (n=6) of emodin and chrysophanol were 99.96% (RSD=0.56%) and 99.29% (RSD=0.32%) respectively. It is believed that the method is simple, reliable, accurate, and repeatable, desirable for the quality control of Shenfu Qiangxin Pill.
