目的:观察经前舒颗粒对郁怒模型大鼠海马5-HT3B受体蛋白表达和mRNA水平以及5-HT含量的影响,研究郁怒情绪的中枢机制,探讨经前舒颗粒的中枢作用机制。方法:以居住入侵方法制备大鼠郁怒情绪模型,给药组给予调肝方药经前舒颗粒,模型组给予灭菌饮用水,并设不造模正常组,用蛋白质印迹技术(WB)检测各组大鼠海马5-HT3B受体蛋白水平表达量,半定量RT-PCR技术检测各组大鼠海马5-HT3B受体mRNA的表达。高效液相色谱-荧光检测器检测海马中5-HT的含量。结果:与正常组比较,模型组大鼠海马5-HT3B受体mRNA水平和蛋白表达水平都极显著升高(P<0.01),5-HT含量显著下降(P<0.05);与模型组比较,经前舒颗粒组大鼠海马蛋白表达和mRNA水平都极显著降低(P<0.01),5-HT含量显著升高。结论:郁怒刺激能显著升高大鼠海马5-HT3B受体蛋白表达和mRNA表达水平并显著降低5-HT的含量,调肝方药经前舒颗粒可能通过下调海马过高的5-HT3B受体蛋白表达和mRNA水平以及上调5-HT含量达到调肝解郁治疗疾病的目的。

Objective: To observe the effects of Jingqianshu granule (JQS) on the content of 5-HT and expression of 5-HT3B receptor in rats’ hippocampus,,investigating the micro-mechanism of anger-in emotinal reaction and exploring the central mechanism of the chinese medicine formula. Methods: The rats were divided into three groups: control, model and administration (JQS).The anger-in rats were prepared using resident-intruder stress.And then we detected the content of 5-HT with HPLC-FLD and the expression levels of 5-HT3B receptor mRNA and protein in rats’ hippocampus by RT-PCR and Western blot method. Results: Compared with the control group, the expression levels of 5-HT3B receptor mRNA and protein of model groups were significant higher in hippocampus (P<0.01), and the content of 5-HT decreased significantly. Compared with model groups, the expression levels of 5-HT3B receptor mRNA and protein of administration groups reduced significantly in hippocampus (P<0.01), but the content of 5-HT incrsased remarkbly. Conclusion: Anger-in emotional reaction can lead to changes of the expression levels of 5-HT3B receptor mRNA and protein and the content of 5-HT and the Chinese medicine formula JQS can decrease the expression levels of 5-HT3B receptor mRNA and protein and increase the content of 5-HT to cure the deseases.
