目的:建立测定大鼠体内血浆和唾液中盐酸川芎嗪含量的HPLC分析方法。方法:采用色谱柱:Diamonsil(R)C18 柱(250mm×4.6mm,5μm);流动相∶甲醇∶水(70∶30,v∶v);柱温:30℃;流速:0.8mL·min-1; 检测波长:295nm。结果:血浆和唾液中盐酸川芎嗪浓度在0.5~50μg·mL-1和0.3~30μg·mL-1范围内相关系数分别为0.9992和0.9999,线性关系良好;盐酸川芎嗪血浆和唾液的相对回收率分别为97.03%~101.19%和99.55%~99.88%;日内、日间精密度的RSD<10%。结论:建立了盐酸川芎嗪唾液样品检测方法以及用HPLC法同时测定大鼠体内血浆和唾液中的含量方法,该方法简便、快速、准确,为盐酸川芎嗪的临床药动学研究提供了新的方法学基础。

This study aimed to establish HPLC determination for TMPH concentration in plasma and saliva of rats. The HPLC was performed on a Diamonsil(R) C18 column (250mm×4.6mm, 5μm) using a mobile phase consisting of methanol-water (70:30, v:v) at 30℃ with the flow rate of 0.8mL· min-1. And the detection wavelength is at 295nm. The result showed that the calibration curve was linear in the range of 0.5- 50.0μg·mL-1 (r=0.9992) in plasma and 0.3-30.0μg·mL-1 (r=0.9999) in saliva for the determination of TMPH. The average recoveries of TMPH in plasma and saliva were 97.03%-101.19% and 99.55%-99.88%, respectively. The intra- and inter-day RSDs were less than 10%. It is concluded that this method is simple, rapid, and accurate. It provides a new methodological basis for the clinical pharmacokinetics study of TMPH.
