目的：比较两种不同功效中药复方对SAM小鼠的治疗差异，为中药复方评价体系提供依据。方法：选用8月龄SAMP8小鼠，分为模型（P）组、六味地黄颗粒（PL组）和通络救脑滴丸（PT组）治疗组。同月龄SAMR1小鼠为对照（N）组。通过水迷宫、避暗实验检测小鼠行为学改变，IH染色观察组织病理学变化，WB检测脑组织APP表达差异。结果：与N组比较，P组及PT组行为学、IH、WB检测指标均出现统计学差异。与P组相比，PL组水迷宫潜伏期缩短，避暗潜伏期延长，海马APP表达减少。结论：六味地黄颗粒可以减少SAMP8小鼠海马APP表达，改善其学习记忆障碍，起到治疗痴呆症的作用，但通络救脑滴丸的疗效不佳。此外，不同中药复方的作用特点有差异，应选择适合的动物模型进行药效评价研究。

This study was aimed to compare the effect of Liu-Wei-Di-Huang (LWDH) and Tong-Luo-Jiu-Nao (TLJN) prescriptions on 8-month-old male SAMP8 mice. Behavioral test, immunohistochemistry and Western blotting were used in the investigation of memory impairment and expression of APP. The results showed that SAMP8 and given prescriptions mice presented spatial learning and memory impairments, decline of neuronal cells and accumulation of APP in the hippocampus. Mice administrated with LWDH presented lower APP levels and perform better than mice administrated with TLJN in behavioral test. It was concluded that LWDH may protect against cognitive deficits by declining APP. However, the therapeutic effects of TLJN on these aspects are not good. Furthermore, the drug action characteristics of different traditional Chinese medicine (TCM) prescriptions are different. Therefore, appropriate animal models should be used to develop and evaluate TCM prescriptions for senile dementia.
