目的:建立高效液相色谱-蒸发光散射检测(HPLC-ELSD)法测定新生脉散液中黄芪甲苷含量。方法:色谱柱:Waters SunfireTM C18(4.6 mm×250 mm,5.0 μm);流动相:乙腈-水(35∶65);流速:1 mL·min-1;柱温:35℃;蒸发光散射检测器参数:漂移管温度100℃,载气流速2.6 mL·min-1。结果:线性范围为0.024~0.240 mg·mL-1 (r=0.9997),平均回收率为99.09%,RSD为1.55%(n=6),重复性RSD为1.22%(n=6),精密度RSD为0.64%(n=6),稳定性RSD为1.61%。结论:本方法操作简便,准确灵敏,可用于新生脉散液的质量控制。

This study was aimed to measure astragoloside Ⅳ content in New Shengmaisan Liquid using HPLCELSD method. Astragoloside Ⅳ were assayed on a Waters SunfireTM C18 (4.6 mm×250 mm, 5.0 μm) column, with the mobile phase composed of Acetonitrile-Water (35: 65). The flow rate was 1 mL/min and the column temperature was 35℃. The drift tube had a temperature of 100℃, with the gas flow being set at 2.6 mL/min. The results showed that the calibration curves were linear ranging from 0.024 to 0.240 mg/mL (r = 0.9997) for astragoloside Ⅳ. The average recovery (n = 6) of astragoloside Ⅳ was 99.09% (RSD = 1.55%). It was concluded that this method is simple, accurate, sensitive and can be used for the quality control of New Shengmaisan Liquid by HPLC-ELSD.
