[摘要]
目的:探求防己与附子配伍减毒的可能机制。方法:非相关文献知识发现工具ARROWSMITH在线软件。结果:防己中的有效成分粉防己碱对包括L-型钙离子通道在内的钙离子通道具有拮抗作用,而L-型钙离子通道也是附子中乌头碱在引发心率失常过程中起关键作用的离子靶点。结论:防己可能通过其有效成分粉防己碱对心肌细胞膜上的L-型钙离子通道进行阻滞,从而对抗附子中乌头碱引起心肌细胞内钙离子增加而导致的心律失常,进而发挥配伍解毒的作用。基于非相关文献知识发现法得到的知识关联结果,不仅能为研究者提供新的研究思路,亦可为科学研究提供更为全面的数据支持。

Software of ARROWSMITH based on the theory of Swanson’s non-correlated literature-bases knowledge discovery was used to study the latent relationship between Radix Stephaniae Tetrandrae and toxicity of Radix Aconiti Lateralis Preparata.Tetrandrine has an antagonistic effect on calcium channel,including L-type calcium channel.And the L-type calcium channel also plays a key role in arrhythmia process triggered by aconitine.It may be the latent mechanism of detoxification in combination of Radix Stephaniae Tetrandrae and Radix Aconiti Lateralis Preparata.The study indicated that non-correlated literature-bases knowledge discovery not only broadened the thinking of scientists but also pr ovided more comprehensive data for scientific research.
