[摘要]
目的:研究四妙勇安汤不同配伍组合对绿原酸、阿魏酸、肉桂酸和哈巴俄苷提取率的影响。方法:将四妙勇安汤拆方后,采用高效液相色谱法,SunFireTM C18色谱柱(4.6mm×150mm,5μm),以乙腈(A)-0.1%磷酸水溶液(B)为流动相,流速1.0mL·min-1,柱温35℃,测定绿原酸、阿魏酸、肉桂酸和哈巴俄苷的含量并分别计算其提取率。结果:测定了原方及拆方后共14组样品中绿原酸、阿魏酸、肉桂酸和哈巴俄苷的含量并计算相应提取率,其回归方程分别为Y=2945611.51X-77211.93(r=0.9996),Y=5266108.02X-2733.36(r=0.9996),Y=8599526.24X-11646.29(r=0.9996)和Y=2822342.06X-1685.64(r=0.9998),线性范围分别为0.237-1.422,0.00976-0.05856,0.01568-0.09408和0.00727-0.0436μg,平均加样回收率分别为99.39%(RSD=1.76%),102.59%(RSD=2.55%),103.60%(RSD=1.07%)和99.32%(RSD=2.93%)。结论:与单味金银花、当归相比,拆方组绿原酸和阿魏酸的提取率均有明显降低,而其全方配伍后的提取率仅低于单味药单煎时提取率;玄参中肉桂酸和哈巴俄苷在全方配伍后提取率最高。本研究有助于揭示四妙勇安汤单味药配伍过程中化学组分变化及配伍作用机制。

This study aimed to investigate the influences of different compatible combinations of Simiao Yong'an decoction(SYD)on the extraction rates of chlorogenic acid,ferulic acid, cinnamic acid and harpagoside,which were determined by HPLC with a SunFireTM C18 analytical column(4.6mm×150mm, 5μm) in the dismantling of SYD.The mobile phase was composed of acetonitrile(A)and aqueousphosphoric acid(0.1%)(B)using a gradient elution.Extractions were performed at 35℃ with a flow rate of 1.0mL·min-1.The chlorogenic acid,ferulic acid,cinnamic acid and harpagoside in fourteen groups from the original prescription and dismantlement were detected and analyzed.As a result,the regression formulations for chlorogenic acid,ferulic acid,cinnamic acid and harpagoside were Y=2945611.51X-77211.93(r=0.9996),Y=5266108.02X-2733.36(r=0.9996), Y=8599 526.24X-11646.29(r=0.9996)and Y=2822342.06X-1685.64(r=0.9998), respectively.The individual linearity of the extracts ranged from 0.237 to 1.422μg,0.00976 to 0.05856μg,0.01568 to 0.09408μg and 0.00727 to 0.0436μg,respectively.The average recoveries of chlorogenic acid,ferulic acid,cinnamic acid and harpagoside were 99.39%(RSD 1.76%),102.59%(RSD 2.55%),103.60%(RSD 1.07%)and 99.32%(RSD 2.93%).Compared with single LoniceraeJaponicae Flos and Angelica sinensis(Oliv.)Diels.,the extraction rates of chlorogenic acid and ferulic acid were obviously reduced in the dismantlement of SYD.Those of the whole prescription were superior to the other groups’,while the extraction rates of cinnamic acid and harpagoside of Scrophularia ningpoensis Hemsl.were the highest in the whole prescription.In conclusion,this study may be beneficial to uncovering the material basis changes behind the compatible mechanisms of SYD.
