[摘要]
目的:优选十味盆安颗粒虎杖、醋延胡索的最佳提取工艺。方法:在单因素考察的基础上,选择乙醇浓度、提取时间、提取次数三个主要因素,以提取液中虎杖苷、延胡索乙素提取率及干膏得率的综合评分为响应值,采用响应面试验优选虎杖、醋延胡索的提取工艺,并对最佳工艺进行工艺验证。结果:最佳提取工艺为10倍量59%乙醇加热回流提取2次,每次95min,经三批工艺验证,虎杖苷、延胡索乙素平均提取率分别为87.80%、94.11%,干膏的平均得膏率为15.00%,综合评分为94.71,SD=0.49%。结论:该工艺采用响应面法优化后,稳定可行,有效成分转移率高,适合产业化生产。

This study aimed to optimize extraction process of Polygoni Cuspidati Rhizoma et Radix and Corydalis Rhizoma from Shiwei Pen'an granule(SWPAG).In the study,ethanol concentration, extraction duration,extraction frequency were set up for main indexes according to the results of single factor experiments;the overall weighted scores of dry cream rate,polydatin and tetrahydropalmatine extraction rates for the response values,while response surface method(RSM)for optimum extraction process.It was found that optimal parameters included hotreflux for twice with ten folds of 59.00% ethanol solution,lasting 95min for each time.Under the optimum conditions,the average dry extract rate was 15.00%,average extraction rates of polydatin and tetrahydropalmatine were 87.80% and 94.11%,respectively;the average dry cream rate was 15.00%,while the weighted composite score was 94.71 with the RSD value of 0.49%.As a conclusion,it was beneficial to industrial production for increasing effective components through the stable and optimum process of RSM.
