目的:本文主要探讨中药复方金复康萃取物对人非小细胞肺癌(Non-Small Cell Lung Cancer,NSCLC)A549细胞增殖和凋亡的影响及其分子机制。方法:本文以人非小细胞肺癌A549细胞为研究对象,应用CCK8试剂盒检测了金复康萃取物对细胞增殖活力的影响,流式细胞技术检测金复康对细胞凋亡诱导作用,实时荧光定量逆转录聚合酶链式反应(Real-time qPCR,RT-qPCR)检测细胞凋亡相关基因的表达。结果:金复康萃取物对A549细胞增殖有较显著抑制作用,并且存在剂量依赖效应。与对照组相比,金复康处理组细胞数量明显减少,伴随着大量细胞间距增大,失去原有形态,DAPI细胞核染色结果显示细胞核受到损伤。流式细胞检测实验显示金复康明显诱导A549细胞凋亡。RT-qPCR检测发现金复康上调凋亡相关基因转录表达水平。结论:金复康通过诱导细胞凋亡抑制A549细胞增殖,这一过程可能与其诱导凋亡相关基因FADD和GADD45a表达上调有关。

[Abstract]
The aim of the study was to explore the effects of JFK on the pr oliferation and apoptosis of human nonsmall cell lung cancer(NSCLC),A549 cells,and its mechanism.In this study,we investigated the anticancer activity and the underlying mechanism of JFK extract on A549 cells.Cell viability analysis was detected using Cell Counting Kit 8(CCK8),while cell apoptosis was analyzed by flow cytometry.The genetic expressions involved in the apoptosis were quantified using RT-qPCR.As a result,JFK extract inhibited the proliferation of A549 cells in a dose-dependent manner.Compared with untreated cells,the number of living cells administered with JFK extract was significantly decreased,while numerous cells were enlarged and became flat.DAPI staining results indicated that nuclear damage of A549 cells were induced by JFK extract.Flow cytometry assay showed that apoptosis occurred in A549 cells after the administration of JFK extract.It was found that the levels of apoptosis associated genes were up-regulated detected by real-time qPCR.In conclusion,it was demonstrated that JFK extract inhibited the proliferation of lung cancer cell by means of cell apoptosis which could be involved in the up-regulation of FADD and GADD45a,the apoptosis associated genes.
