[摘要]
目的:观察胶原诱导关节炎(Collagen Induced Arthritis, CIA)小鼠发病高峰期重要脏器的病理变化。方法:20只雄性DBA/1小鼠随机分为正常组和模型组,模型组给予牛II型胶原混合完全弗氏佐剂诱导CIA模型。造模后每周进行关节炎指数评分,于第63天处死小鼠,取心、肝、脾、肺、肾各组织和踝关节,利用HE染色法观察各脏器的病理变化,并采用Real-time PCR法检测小鼠肺组织中转化生长因子β1(Transforming Growth Factor Beta 1, TGF-β1)以及肿瘤坏死因子α(Tumor Necrosis Factor Alpha, TNF-α)的mRNA表达水平。结果:成功建立了DBA/1小鼠胶原诱导关节炎模型。HE染色结果表明,除了踝关节出现明显的病理变化外,CIA小鼠的肺部有大量的炎细胞浸润,肺泡间隔增厚,肺泡腔中有炎性浸出物,而心脏、肝脏、脾脏和肾脏没有出现明显的病理变化。Real-time PCR结果显示病变的小鼠肺组织中TGF-β1和TNF-α的含量均显著高于正常的肺组织。结论:胶原诱导的DBA/1小鼠,其踝关节呈现严重的病理变化,其肺部也出现明显的炎症,且与类风湿关节炎肺部并发症的病理变化有相似之处,这些结果提示该模型可用于类风湿关节炎肺部病变的相关研究。

Objective To investigate the pathological changes of key organs in collagen induced arthritis (CIA) mice. Methods Twenty male DBA/1 mice were grouped into normal group and model group randomly. Mice in model group were induced with bovine type II collagen and complete Freund??s adjuvant. Arthritis index was scored once every week after immunization. The mice were sacrificed at Day 63 and the heart, liver, spleen, lung, kidney and ankle joint were collected to observe their pathological changes by H&E staining. In addition, the mRNA levels of transforming growth factor beta 1 (TGF-β1) and tumor necrosis factor alpha (TNF-α) in the lung were detected by Real-time PCR. Results CIA model was successfully established in DBA/1 mouse. H&E staining analysis showed massive inflammatory cells infiltration, thickened alveoli septum in lungs except for apparent pathological changes in ankle joints of CIA mice. There was no obvious pathological changes in hearts, livers, spleens and kidneys of CIA mice. Real-time PCR results indicated that mRNA levels of TGF-β1 and TNF-α in the lungs with pathological changes were significantly higher than those in normal lungs. Conclusions DBA/1 mice with CIA not only showed severe pathological changes in ankle joints, but also obvious inflammatory responses in lung. These changes in lung of CIA mice were similar to those in rheumatoid arthritis patients. These results indicated that DBA/1 mice with CIA could be used for studies related to lung disease associated with rheumatoid arthritis.
