膜蛋白是细胞膜功能的主要承担者,参与信息传递、物质交换、催化反应等生物学功能,也是60%-70%药物的主要作用靶点。正是基于膜蛋白的重要性,以研究整体膜蛋白水平的膜蛋白组学(membrane proteomics)逐渐兴起,并发展成为蛋白质组学研究生物学功能中的重要组成部分。由于膜蛋白的疏水性、低丰 度等特点,在进行Shotgun质谱分析前仍需要对样品进行一些特殊的处理,本文针对膜蛋白样品的前处理及相 关应用研究的进展进行了综述。

[Abstract]
Membrane protein (PMs) plays a key role in the biological function of plasma membrane, and takes part in information transmission, material exchange, catalytic reaction, etc. More importantly, many PMs are the targets for about 60%-70% approved drugs. Because of the importance of PMs, more and more researchers are focusing on the membrane proteomics which can simultaneously reflect the change of thousands of proteins. As a part of proteomics, however, membrane proteomics have some difficulties in sample preparation prior to LC-MS analysis because the PMs are lowabundance and high-hydrophobicity. In the review, we aimed at the advances of sample preparation and the applications for PMs in recent years.
