[摘要]
目的 建立高效液相色谱法(high performance liquid chromatography,HPLC)测定四磨汤中4种槟榔生物碱的含量,为四磨汤质量控制提供技术支撑和数据支持。方法 采用高效液相色谱二极管阵列检测器联用仪(high performance liquid chromatography-diode array detector,HPLC-DAD)对槟榔生物碱进行含量测定,色谱柱:阳离子Agela Venusil SCX(T) (4.6 mm× 250 mm, 5 μm)柱,流动相:乙腈: 0.5%磷酸(三乙胺调pH值2.40)=45:55,流速:1.0 mL·min-1,检测波长:215 nm,柱温:25℃。结果 去甲基槟榔次碱、盐酸槟榔次碱、盐酸去甲基槟榔碱和氢溴酸槟榔碱在各自的浓度范围11.94-95.42、19.96-159.68、7.928-63.424、19.94-159.52 μg·mL-1内线性关系良好,相关系数R2均为0.999,检测限(limit of detection,LOD)分别为0.16、0.12、0.01、0.47 μg·mL-1,加样回收率分别为101.04%、101.263%、103.45%和98.43%。6份四磨汤样品中去甲基槟榔次碱、槟榔次碱、去甲基槟榔碱和槟榔碱的平均含量分别为0.039 3、0.059 0、0.002 5、0.088 8 mg·mL-1,且去甲基槟榔次碱和槟榔次碱含量波动比较大。结论 本方法灵敏性好、准确度高,简便可靠,可用于四磨汤中4种槟榔生物碱含量的同时测定及质量控制。

[Abstract]
Objective To establish a high performance liquid chromatography (HPLC) method for simultaneous determination of four areca alkaloids in Simo decoction. It provides technical and data supports for quality control of Simo decoction.Methods The contents of areca alkaloids were determined by high performance liquid chromatography-diode array detector (HPLC-DAD). The separation of areca alkaloids was performed on column which was an Agela Venusil SCX(T) chromatographic column (4.6 mm × 250 mm, 5 μm) and the mobile phase was consisted of acetonitrile-0.5% phosphoric acid (adjusted with triethylamine to pH = 2.40) at 45:55 at the flow rate of 1.0 mL·min-1. The detection wavelength was set at 215 nm with the column temperature of 25℃.Results The linear ranges of guvacine, arecaidine hydrochloric, guvacoline hydrochloride and arecoline hydrobromide were 11.94-95.42, 19.96-159.68, 7.928-63.424, 19.94-159.52 μg·mL-1, respectively, with R2 of 0.999. Limit of detection (LOD) were 0.16, 0.12, 0.01, 0.47μg·mL-1, and the recovery rates were 101.04%, 101.263%, 103.45% and 98.43%. The detected average content of guvacine, arecaidine, guvacoline and arecoline were 0.039 3, 0.059 0, 0.002 5, 0.088 8 mg·mL-1 in 6 copies the Simo decoction. And the content of guvacine and arecaidine fluctuated greatly.Conclusion The established method is simple, reliable and accurate, which can be used for the determination of four areca alkaloids, further for the quality control of Simo decoction.
