[摘要]
目的 本研究遴选中医培土生金法的代表方参苓白术散合麦门冬汤加减方灌胃治疗染矽尘肺纤维化(肺脾两虚型)大鼠,观察中药治疗各组对染矽尘大鼠肺组织HMGB1、IL-6、PDGF、K-Ras蛋白含量,基因表达水平作用的影响改变。方法 对健康SD大鼠随机分为正常对照组和造模组。造模组分别给予大黄煎液灌服造模法与二氧化硅(SiO2)气管滴注诱导染矽尘肺纤维化造模法结合,制造染矽尘肺纤维化(肺脾两虚型)模型,待造模成功后,造模组随机分为5组,分别给予生理盐水、激素、中药高剂量、中药中剂量和中药低剂量进行治疗。各组动物于造模成功后30 d处死。ELISA检测各组肺组织高迁移率族蛋白1(HMGB1)、白介素6(IL-6)、血小板衍生因子(PDGF)、K-RAS蛋白含量并提取各组肺组织RNA,采用反转录合成cDNA,通过RT-PCR技术扩增HMGB1、IL-6、PDGF、K-RAS基因。结果 中药各组肺组织HMGB1、IL-6、PDGF、K-Ras蛋白含量,基因表达水平明显低于染矽尘组(P < 0.05),但中药各组肺组织HMGB1、IL-6、PDGF、K-Ras蛋白含量,基因表达水平明显高于对照组(P < 0.05)。结论 中药各组能显著降低肺组织HMGB1、IL-6、PDGF、K-Ras蛋白含量和基因表达,HMGB1与PDGF间存在正相关关系。

[Abstract]
Objective To observe the effect of Shenling Baizhu Powder and Maimendong Decoction on HMGB1, IL-6, PDGF, K-Ras protein content and gene expression in lung tissue of Silicosis Rats Treated with traditional Chinese medicine Change.Methods Healthy SD rats were randomly divided into normal control group and model group. Modeling group were given Rhubarb decoction perfusion modeling method and silica dioxide (SiO2) tracheal drip induced silicosis fibrosis modeling method, silicosis pneumoconiosis fibrosis (deficiency of both lung and spleen) model, after the success of modeling, modeling group was randomly divided into five groups, were given normal saline, hormones, high doses of traditional Chinese medicine, medium doses of traditional Chinese medicine, low doses of traditional Chinese medicine. All animals were sacrificed at 30d after successful modeling. High mobility group 1 (HMGB1), interleukin-6 (IL-6), platelet-derived factor (PDGF), K-RAS protein were detected by ELISA, and RNA was extracted from lung tissues of each group. The genes of HMGB1, IL-6, PDGF and K-RAS were amplified by RT-PCR.Results The levels of HMGB1, IL-6, PDGF, K-Ras protein and gene expression in lung tissues of TCM groups were significantly lower than those of silica dust group (P < 0.05), but the levels of HMGB1, IL-6, PDGF and K-Ras protein in lung tissues of TCM groups were significantly higher than those of control group (P < 0.05).Conclusion The contents and gene expression of HMGB1, IL-6, PDGF, K-Ras protein in lung tissues were significantly decreased in each group. There was a positive correlation between HMGB1 and PDGF.
