[摘要]
目的 基于高效液相色谱法指纹图谱(High performance liquid chromatography,HPLC)、相似度评价法、主成分分析法对不同产地、不同生长方式喜马拉雅紫茉莉的品质差异进行研究。方法 通过HPLC梯度洗脱筛选不同产地、不同生长方式喜马拉雅紫茉莉最佳检测条件、建立指纹图谱。以指纹图谱中不同产地、不同生长方式喜马拉雅紫茉莉的共有成分的含量为变量,运用SIMCA 13.0软件和中药色谱指纹图谱相似度评价系统(2004)A版进行PCA分析和相似度评价;结果 筛选得到最佳实验条件如下:提取溶剂为甲醇、系统4、检测波长280 nm、进样量为20 μL、柱温25℃,由此建立了喜马拉雅紫茉莉HPLC指纹图谱共有系统,鉴定得到9个共有指纹峰;同时,不同产地、不同生长方式喜马拉雅紫茉莉中有17个样本的相似度在0.90以上;主成分分析结果显示:野生品种和栽培品种分布于图的左右两侧,存在显著性差异。结论 本研究建立的喜马拉雅紫茉莉HPLC指纹图谱结合相似度和PCA分析,能够为喜马拉雅紫茉莉的品质提供客观、有效的分析。

[Abstract]
Objective To establish the HPLC fingerprint of Mirabilis himalaica (Edgew.) Heim. and analyze the quality of Mirabilis himalaica (Edgew.) Heim. samples from different habitats and under different growth modes. Methods The experimental conditions of high performance liquid chromatography was screened and optimized, including organic solvent of pretreatment and extraction, elution conditions, detection wavelength, sampling dosage and column temperature. Results The optimum condition appeared as follows: extraction solvent was methanol, system 4, detection wavelength was 280 nm, injection amount was 20 μL, and column temperature was 25℃. 9 common fingerprint peaks were targeted and identified. Afterwards, principal component analysis (PCA) showed that difference was significant among samples under different growth patterns, namely wild and cultivated. Conclusion The HPLC fingerprint of Mirabilis himalaica (Edgew.) Heim. established in this study can provide an important reference value for the quality evaluation of Mirabilis himalaica (Edgew.) Heim. Also it gives insights to further research on quality control of traditional Chinese medicine under growth patterns.
