目的 对野菠萝提取物的降脂减肥作用进行药效学评价。方法 7周龄SPF级雄性C57BL/6小鼠,高脂高糖饲料喂养诱导肥胖模型,正常饲料喂养正常对照组小鼠。喂养30天后,筛选肥胖模型小鼠随机分为模型组、野菠萝提取物(YBL)高、中、低剂量组,分别给予蒸馏水与2.4 mg/mL、1.2 mg/mL、0.6 mg/mL的YBL溶液代水饮54天(相当于每天给药YBL剂量约400 mg/kg、200 mg/kg、100 mg/kg)。实验期间每周记录动物体重、摄食量变化。实验结束时处死动物,称重脂肪并计算脂肪/体重比值、体重增重;采血检测动物血清中总胆固醇(CHO)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL)、低密度脂蛋白胆固醇(LDL)和葡萄糖(Glu)水平;取肝脏制作苏木精-伊红染色法(hematoxylin-eosin staining,HE)染色病理切片,观察肝组织脂肪变情况。结果 肥胖模型组动物体重、增重、脂肪/体重比值、血清脂质水平均显著高于正常对照组(p <0.01~0.05)。2.4 mg/mL、1.2 mg/mL的YBL溶液代水饮干预组可显著降低小鼠体重增加趋势,减少最终的增重(p < 0.01-0.05),降低血清CHO、LDL、Glu水平(p < 0.01-0.05),减轻肝组织脂肪变程度。结论 野菠萝提取物可以有效抑制高脂高糖饲料喂养诱导的小鼠肥胖,降低血清CHO、LDL、Glu水平,改善肝脏组织病理形态。

[Abstract]
Objective To evaluate the effect of Pandanus tectorius on weight loss and blood lipid-lowering in mice.Methods 7weeks old SPF male C57BL/6 mice were divided into normal group, high-fat high-sugar fed group. Mice in the normal group were fed ordinary fodder and others were fed high-fat high-sugar fodder. After 30 days, the mice in high-fat high-sugar fed group were selective divided into obesity model group and Pandanus tectorius fed group (drink three different concentration solutions instead of water, that"s equivalent to about 400 mg/kg, 200 mg/kg and 100 mg/kg of Pandanus tectorius extract per day).There mice raised for another 54 days. Body weights and food intakes were determined every week. At the end of the experiment, the body fat were weighted and the fat/body weight, body weight gains were calculated. The contents of CHO, TG, HDL, LDL and glucose in serum were measured. The liver tissue pathological slices by HE staining were also observed.Results Body weight, weight gain, fat/body weight ratio and serum lipid level in the obesity model group were significantly higher than those in the normal control group (p < 0.01-0.05).The intervention group with 2.4 mg/mL and 1.2 mg/mL YBL solution could significantly reduce the weight gain trend of mice, reduce the final weight gain (p < 0.01~0.05), reduce the serum CHO, LDL and glucose levels (p < 0.01-0.05), and reduce the degree of liver tissue fat change.Conclusions Pandanus tectorius extract can effectively inhibit obesity in mice induced by high-fat and high-sugar diets, reduce serum CHO, LDL and glucose levels, and improve the pathological morphology of liver tissues.
