[摘要]
目的 研究不同时间针刺足三里穴对炎性痛症大鼠不同部位中瞬态电压感受器阳离子通道香草酸受体亚型1(TRPV1)的影响。方法 使用清洁级雄性SD大鼠54只,驯养7天后,经过初选后,将确定出来的实验对象随机分为待模型复制组和空白组,然后将模型复制成功后的实验对象分为模型组和模针组,将模型组、模针组和空白组分别在特定的两个时间点(ZT8、ZT16)进行相应地处理;在第7天针刺治疗结束后,立即检测相对应时间点大鼠的痛阈,选择免疫组织化学检测法来确定提取的大鼠穴区局部皮肤、腰椎4-6的脊髓及背根神经节样本中TRPV1的情况。结果 ①在穴区皮肤:ZT8时间点,模型组皮肤组织中TRPV1含量明显多于空白组(P < 0.05);而模针组穴区皮肤样本中TRPV1含量明显低于模型组(P < 0.05)。相应地,在ZT16时间点,模型组皮肤样本目标中TRPV1的表达亦明显多于空白组(P < 0.05);而类似地,模针组穴区皮肤样本中TRPV1的表达则少于模型组(P < 0.05)。②在脊髓中:ZT8时间点,模型组脊髓节段中TRPV1的含量明显多于空白组中TRPV1的表达量(P < 0.01),而与模型组比较,模针组脊髓节段样本组织中目标对象TRPV1被上调的更为显著(P < 0.01);在ZT16时,模型组脊髓节段中TRPV1的含量与空白组脊髓节段中的TRPV1相比显著增加(P < 0.01),与模型组相比,模针组脊髓节段中TRPV1的表达量明显下调(P < 0.05)。③在背根神经节中:ZT8与ZT16两个时间点,模型组DRG样本中TRPV1含量皆明显多于空白组(P < 0.01),而模针组DRG样本中TRPV1的表达则低于模型组(P < 0.05)。结论 ①针刺足三里穴能够调节炎性痛症大鼠多个层面组织中TRPV1的含量。②不同时间点针刺镇痛效应差异可能与不同时间点针刺后各个组织中TRPV1表达的差异有关。

[Abstract]
Objective To study the effects of acupuncture at Zusanli on transient voltage receptor cation channel vanilloid receptor subtype 1 (TRPV1) in different parts of rats with inflammatory pain.Methods 54 clean male SD rats were acclimated for 7 days, and then screened according to the basic pain threshold. The selected experimental rats were randomly divided into model and control group. Rats with successful modeling were randomly divided into model group and model needle group according to the corresponding basic pain thresholds. Each group was divided into two subgroups of ZT8 and ZT16 according to different treatment time, and 9 rats in each group. On the 7th day after the completion of the acupuncture treatment, the pain threshold of the rats at the corresponding time points was immediately detected, and then the local skin of the acupuncture points, the spinal cord of the lumbar vertebrae 4-6 and the dorsal root ganglia were extracted, and the expresssion of TRPV1 in local skin,dorsal root ganglion and spinal cord in the rat acupoint area were detected by immunohistochemisty.Results ① In the skin: at the ZT8 time point, the content of TRPV1 in the skin of the model group was significantly higher than that of the control group(P < 0.05); while the model group was compared with the needle group, the expression of TRPV1 in the model group was Significantly reduced (P < 0.05). Correspondingly, at the ZT16 time point, compared with the control group, the content of TRPV1 in the skin of the model group was significantly increased (P < 0.05); while the content of TRPV1 in the skin of the model needle group was significantly decreased than that of the model group(P < 0.05). ② In the spinal cord: at the ZT8 time point, the content of TRPV1 in the model group was significantly higher than that in the control group(P < 0.01), while the expression level of TRPV1 in the model needle group was significantly decreased compared with the model group (P < 0.01). At the ZT16 time point, the expression of TRPV1 in the model group was significantly increased compared with the control goup (P < 0.01), and the expression level of TRPV1 in the model needle group was significantly lower than that in the model group (P < 0.05). ③ In the dorsal root ganglia (DRG): at the ZT8 time point,the content of TRPV1 in the model group was significantly higher than that in the control group(P < 0.01), while the expression of TRPV1 in the model needle group was significantly lower than that in the model group(P < 0.01); At the ZT16 time point, compared with the control group, the expression of TRPV1 in the model group was significantly increased (P < 0.01), while the expression of TRPV1 was significantly decreased in the model needle group compared with the model group(P < 0.05).Conclusion ① Acupuncture at Zusanli can regulate the content of TRPV1 protein in local skin, spinal cord and dorsal root ganglion of rats with inflammatory pain. ②The analgesic effect of acupuncture at different time points may be related to the difference of TRPV1 expression in local skin, spinal cord and dorsal root ganglion after acupuncture at different time points.
