目的 探究两种剂型龟鹿二仙胶对于腺嘌呤诱导肾阳虚型少弱精子症大鼠的干预作用。方法 32只8周龄SD大鼠,体质量200 g-220 g,随机分4组,分别为空白组(Control)、模型组(Model)、龟鹿二仙胶饮片膏组(GLEX_T)、龟鹿二仙胶颗粒膏组(GLEX_G),每组8只;空白组灌胃等体积蒸馏水,其余4组给与腺嘌呤混悬液灌胃,连续灌胃30天;从31天起,空白组及模型组灌胃等体积蒸馏水,龟鹿二仙胶饮片膏组予龟鹿二仙胶饮片膏灌胃,龟鹿二仙胶颗粒膏组予龟鹿二仙胶颗粒膏灌胃,连续灌胃至60天。每日灌胃剂量10 mL?kg-1。给药完毕后处死,检测血清性激素(T、E2、FSH、LH)、精液参数(精子活力、活率、密度)和睾丸HE染色实验。结果 龟鹿二仙胶饮片膏组与龟鹿二仙胶颗粒膏组都能调节性激素分泌、改善大鼠精液参数(P < 0.01,P < 0.05),且在干预效果上无显著差异(P > 0.05);龟鹿二仙胶饮片膏组与龟鹿二仙胶颗粒膏都对大鼠睾丸组织有保护作用。结论 两种剂型龟鹿二仙胶对于腺嘌呤诱导肾阳虚型少弱精子症大鼠有干预作用且疗效相近,可能与其对下丘脑-垂体-睾丸轴的调控、内分泌激素的改变,睾丸组织的保护有关。

Objective To explore the intervention effect of two kinds of Guilu Erxian gum on oligoasthenospermia rats with kidney yang deficiency induced by adenine.Methods A total of 32 Sprague Dawley rats, 8 weeks old of age weighing 200 g-220 g, were randomly divided into four groups: the blank group, model group, Guilu Erxian gum traditional ointment (GLEX_T) group and Guilu Erxian gum granule ointment (GLEX_G) group, with 8 rats in each group. The blank group was given an equal volume of distilled water, and the other four groups were given adenine suspension by gavage for 30 days. From 31 days on, the blank group and model group were given an equal volume of distilled water, the GLEX_T group was given Guilu Erxian gum traditional ointment, and the GLEX_G group was given Guilu Erxian gum granule ointment by gavage for 60 days. The daily dose was 10 mL?kg-1. After treatment, serum, testis, epididymis and spermatozoa were extracted, serum sex hormone levels were detected by ELISA, spermatozoa were detected by CASA, morphological changes of testis were observed by HE staining.Results Both GLEX_T and GLEX_G could regulate the secretion of sex hormone and improve the semen parameters of rats (P < 0.01, P < 0.05), and there was no significant difference in the intervention effect (P > 0.05); both Guilu Erxian gum traditional ointment and Guilu Erxian gum granule ointment had a protective effect on testicular tissue of rats.Conclusion The two formulations of Guilu Erxian gum have similar effects on adenine-induced oligoasthenospermia rats with kidney yang deficiency, which may be related to the regulation of the hypothalamus-pituitary testicular axis, the change of endocrine hormones and the protection of testicular tissue.
