目的 考察弩药液对膝骨性关节炎大鼠关节软骨组织的改变及关节液中炎症因子IL-1β、MMP-3、TNF-α含量的影响,并探讨其抗炎作用可能的机制。方法 取适应性饲养后的大鼠随机分为空白组、模型组、阳性组、弩药高、中、低剂量组。用4%木瓜蛋白酶对(空白组除外)各组大鼠进行膝骨性关节炎造模。造模成功后,在大鼠左后腿膝关节处,空白组、模型组用50%乙醇涂抹,其余组涂抹相应药物。每天给药1次,连续30天。末次给药24 h后,取膝关节软骨组织,观察其病理学变化情况,并进行Mankin,s评分;取关节液,采用酶联免疫吸附测定法检测关节液中IL-1β、MMP-3、TNF-α的含量;用熵权法计算综合评分,以此评价弩药对膝骨性关节炎模型大鼠上述指标的综合影响。结果 与空白组比,模型组Mankin,s评分及炎症因子含量显著升高(P < 0.05)。与模型组比,阳性组和弩药高、中、低剂量组Mankin,s评分及炎症因子含量均明显降低(P < 0.05)。熵权法计算得各给药组综合评分分别为0.2401(阳性组)、0.3327(弩药高剂量组)、0.2259(弩药中剂量组)、0.2013(弩药低剂量组)。结论 弩药对木瓜蛋白酶诱导的膝骨性关节炎大鼠有一定治疗作用,且药物高剂量组作用强于中、低剂量组,其作用机制可能与改善大鼠膝关节软骨细胞活性及抑制大鼠关节液中IL-1β、MMP-3、TNF-α的表达有关。

Objective To investigate the effect of crossbow medicinal solution on the changes of articular cartilage tissue and the content of inflammatory factors IL-1β, MMP-3 and TNF-α in the joint fluid of rats with knee osteoarthritis, and to explore the possible mechanism of its anti-inflammatory effect.Methods The rats were randomly divided into blank group, model group, positive group, and high, medium and low dose groups of crossbow medicine after adaptive feeding. The rats in each group (except the blank group) were modeled with 4% papain for osteoarthritis of knee. After successful modeling, at the knee joint of the left hind leg of the rats, the blank and model groups were coated with 50% ethanol, and the remaining groups were coated with the corresponding drugs. The drug was administered once a day for 30 days. After 24 h of the last administration, the cartilage tissue of the knee joint was taken to observe the pathological changes and Mankin, s score; the joint fluid was taken and the contents of IL-1β, MMP-3 and TNF-α in the joint fluid were detected by enzyme-linked immunosorbent assay; the comprehensive score was calculated by entropy weighting method, which was used to evaluate the comprehensive effect of the crossbow drug on the above indexes in the knee osteoarthritis model rats.Results Mankin''s scores and inflammatory factor content were found to be significantly higher in the model group compared with the blank group (P < 0.05). Compared with the model group, Mankin''s score and inflammatory factor content were significantly lower in the positive group and the high, medium and low dose groups of the crossbow medicine (P < 0.05). The entropy weighting method calculated the composite score of each administration group as 0.2401 (positive group), 0.3327 (high dose group of crossbow drug), 0.2259 (medium dose group of crossbow medicine), and 0.2013 (low dose group of crossbow medicine), respectively.Conclusion The crossbow medicine solution has a certain therapeutic effect on rats with knee osteoarthritis induced by papain, and the high-dose drug group was stronger effect than the middle and low-dose groups. Its mechanism of action may be related to the improvement of knee chondrocyte activity of rats and inhibition the expression of IL-1β, MMP-3 and TNF-α in the synovial fluid of rats.
