[摘要]
目的 探究三七粉发挥消肿定痛功效的质量标志物。方法 首先,采用小鼠醋酸扭体实验,将ICR小鼠随机分成模型对照组、阳性对照阿司匹林组(0.12 g·kg-1)、三七粉低(1.1 g·kg-1)、中(2.2 g·kg-1)、高(4.4 g·kg-1)剂量组、三七总皂苷低(0.25 g·kg-1)、中(0.5 g·kg-1)、高(1.0 g·kg-1)剂量组,连续给药3天每日1次,第3天给药1 h后腹腔注射1%醋酸溶液,记录15 min内扭体次数。进一步采用LPS诱导的小鼠腹腔巨噬细胞RAW264.7炎症模型,探讨三七粉中的三七总皂苷及14个代表性单体成分对细胞上清液中NO、TNF-α和IL-6的含量影响。结果 实验表明,与模型组比较,三七粉及三七总皂苷中、高剂量组均能显著减少扭体反应次数(P<0.05,P<0.01);三七总皂苷及14个化合物均能显著降低细胞上清液中NO、TNF-α、IL-6含量(P<0.05)。结论 三七粉具有良好的抗炎、镇痛作用,其发挥消肿定痛的药效物质基础可能为三七皂苷R1、人参皂苷Re、人参皂苷Rg1、人参皂苷Rg2、人参皂苷Rh1、人参皂苷Rd、人参皂苷Rg3、人参皂苷Rb1、人参皂苷Rb2、人参皂苷Rb3、人参皂苷F2、人参皂苷Rk1、槲皮素、三七素。

[Abstract]
Objective To explore the quality markers of Panax Notoginseng powder for reducing swelling and relieving pain.Methods ICR mice were randomly divided into model control group, positive control aspirin group (0.12 g·kg-1), low (1.1 g·kg-1), medium (2.2 g·kg-1) and high (4.4 g·kg-1) dosage groups of Panax Notoginseng powder, and low (1.1 g·kg-1), medium (2.2 g·kg-1) and high (4.4 g·kg-1) dosage groups of Panax Notoginseng saponins. The mice were administered once a day for 3 days, and 1% acetic acid solution was injected intraperitoneally 1 hour after administration on the third day. The twisting times in 15 minutes were recorded. Furthermore, LPS-induced mouse peritoneal macrophage RAW264.7 inflammatory model was used to investigate the effects of total saponins and 14 representative compounds in Panax Notoginseng powder on the contents of NO, TNF-α and IL-6 in cell supernatant.Results Compared with the model group, medium and high dose groups of Panax Notoginseng powder and Panax Notoginseng saponins medium could significantly reduce the number of writhing reactions (P<0.05, P<0.01). The Panax Notoginseng saponins and 14 representative compounds could significantly reduce the content of NO, TNF-α and IL-6 in cell supernatant (P<0.05).Conclusion Panax Notoginseng powder has good anti-inflammatory and analgesic effects. The material basis of its reducing swelling and relieving pain effect may be notoginsenoside R1, ginsenoside Re, ginsenoside Rg1, ginsenoside Rg2, ginsenoside Rh1, ginsenoside Rd, ginsenoside Rg3, ginsenoside Rb1, ginsenoside Rb2, ginsenoside RB3, ginsenoside F2, ginsenoside RK1, quercetin and dencichine.
