[摘要]
目的 探讨丁香酚包合物对心肌缺血再灌注损伤大鼠心脏的保护作用及机制研究。方法 将丁香酚与无水乙醇溶液按照(1:3)的比例混合互溶备用,采用超声法制备丁香酚包合物;将 Wistar雄性大鼠,采用随机方法分为假手术组、模型组、包合物基质组、地尔硫?组、丁香酚包合物低、中、高剂量组,每组10只。假手术组、模型组灌胃CMC-Na,包合物基质组灌胃β-环糊精CMC-Na混悬液,地尔硫?组灌胃地尔硫?CMC-Na混悬液,丁香酚包合物低、中、高剂量组分别按0.125 g/kg/天、0.25 g/kg/天、0.5g/kg/天(含有实际丁香酚含量95 mg、190.5 mg、381 mg)灌胃CMC-Na混悬液,每天给药1次,连续给药15天。末次给药1 h后,在大鼠的冠状动脉左前降支处结扎30 min,恢复再灌注24 h建立MIRI模型;采用BL-420S实验系统检测大鼠心电信号;采用全自动生化仪检测大鼠血清中肌酸激酶(CK)、肌酸激酶同工酶(CK-MB)、乳酸脱氢酶(LDH)和谷草转氨酶(AST)的活力;采用试剂盒法测定大鼠心肌组织中丙二醛(MDA)和超氧化物歧化酶(SOD)的活力;采用TTC染色法检测大鼠心肌梗死面积;采用蛋白免疫印迹法(Western blotting)法检测大鼠心肌缺血组织中GLUT4、cTn1、P-AMPK/AMPK的蛋白表达水平。结果 与模型组比较,丁香酚包合物高剂量组可显著降低大鼠血清中LDH、CK、CK-MB、AST的水平,(P<0.01),显著增加大鼠心肌组织中SOD的水平(P<0.01),显著降低大鼠心肌组织中MDA的水平(P<0.01),显著下调P-AMPK/AMPK的蛋白表达含量(P<0.01),显著减少cTn1蛋白表达的含量(P<0.01),显著增加GLUT4蛋白表达水平(P<0.01)。结论 丁香酚包合物可缓解心肌缺血再灌注损伤,其保护作用可能与抑制磷酸化AMPK的表达以及cTn1的表达有关。

Objective To explore the protective effect and mechanism of eugenol inclusion compound on the heart of myocardial ischemia-reperfusion injury in rats.Methods Wistar male rats were randomly divided into sham operation group, model group, and package group. Compound matrix group, diltiazem group, and eugenol inclusion compound low, medium and high dose groups, with 10 rats in each group. The blank group and the model group were intragastrically administered with CMC-Na, the inclusion compound matrix group was intragastrically administered with β-cyclodextrin CMC-Na suspension, and the diltiazem group was intragastrically administered with diltiazem CMC-Na suspension and eugenol. The low, medium and high dose groups of the inclusion compound were calculated according to 0.125 g/kg/d, 0.25 g/kg/d, 0.5 g/kg/d (contains the actual eugenol content 95 mg, 190.5 mg, 381 mg) CMC-Na suspension was intragastrically administered once a day for 15 consecutive days. One hour after the last administration, the rat''s left anterior descending coronary artery was ligated for 30 minutes and reperfusion was restored for 24 hours to establish the MIRI model; the BL-420S experimental system was used to detect the rat''s ECG signal; the automatic biochemical analyzer was used to detect the rat serum of the activity of creatine kinase (CK), creatine kinase isoenzyme (CK-MB), lactate dehydrogenase (LDH) and aspartate aminotransferase (AST); the kit method was used to determine malondialdehyde (MDA) and superoxide dismutase (SOD) activity; TTC staining method was used to detect rat myocardial infarction area; Western blotting method was used to detect GLUT4, cTn1, P-AMPK/AMPK in rat myocardial ischemic tissue/AMPK protein expression level.Results Compared with the model group, the high-dose eugenol inclusion compound group can significantly reduce the levels of LDH, CK, CK-MB, and AST in rat serum (P<0.01), and significantly increase the level of SOD in rat myocardial tissue (P<0.01), significantly reduce the level of MDA in rat myocardial tissue (P<0.01), significantly decrease the protein expression content of P-AMPK/AMPK (P<0.01), and significantly reduce the protein expression content of cTn1 (P<0.01), significantly increased the expression level of GLUT4 protein (P<0.01).Conclusion Eugenol inclusion compound can alleviate myocardial ischemia-reperfusion injury, and its protective effect may be related to the inhibition of the expression of phosphorylated AMPK and the expression of cTn1.
