目的 通过高脂饮食加4-乙烯基环己烯二环氧化物(VCD)药物诱导小鼠卵巢功能衰退的方法,建立围绝经期肥胖动物模型,为中医药领域开展围绝经期肥胖相关实验研究提供可靠的模型基础。方法 8周龄雌性C57BL/6N小鼠20只,随机分为空白组,4-乙烯基环己烯二环氧化物组,每组10只,观察两组小鼠动情周期变化、体质量、体长、腰围、Lee’s指数、脂肪湿重、采用酶联免疫吸附测定检测血清雌激素、促卵泡刺激素(FSH)水平,比较两组小鼠体重、血清性激素水平的变化,比较两组卵巢大小、湿重及组织形态学变化。结果 与空白组比较,模型组动情周期紊乱,卵巢组织萎缩,卵泡减少,体质量、体长、腰围、Lee’s指数、脂肪湿重均大于空白组,均具有统计学差异(P<0.05)。结论 使用高脂饮食喂养加4-乙烯基环己烯二环氧化物诱导的方法可以导致小鼠卵巢功能衰退、体重增加,是一种围绝经期肥胖的理想动物模型,该模型较为稳定,适合用于中医药领域对围绝经期肥胖的基础研究。

Objective To establish a perimenopausal obesity mice model by using a high-fat diet (HFD) combine with 4-vinylcyclohexene diepoxide (VCD) drug to induce ovarian function decline in mice, and to develop perimenopause for the field of traditional Chinese medicine obesity-related experimental research and provide a reliable model basis.Methods Twenty female C57BL/6N mice aged 8 weeks were randomly divided into blank group and VCD group, with 10 mice in each group. The changes of estrous cycle, body weight, body length, waist circumference, Lee''s index and fat wetness of the two groups of mice were observed. Enzyme-linked immunosorbent assay (ELISA) was used to detect serum estrogen (E2) and follicle stimulating hormone (FSH) levels. The body weight and serum sex hormone levels of the two groups of mice were compared. The changes in body weight and serum sex hormone levels of the two groups of mice were compared, and the size, wet weight and histomorphological changes of the ovaries were compared between the two groups.Results Compared with the blank group, the estrous cycle of the model group was disordered, the ovarian tissue was atrophied, and the follicles were reduced. The body weight, body length, waist circumference, Lee''s index, and wet fat weight of the model group were larger than those of the blank group, all of which were statistically different (P<0.05).Conclusion The method of using HFD combine with VCD induction could lead to the decline of ovarian function and weight gain in mice. It is an ideal mice model for perimenopausal obesity.
