[摘要]
目的:基于UHPLC-Q-Exactive-Orbitrap-MS/MS对糖尿灵片中的化学成分进行定性分析。方法:使用Waters ACQUITY HSS T3色谱柱(100 mm×2.1 mm,1.8 μm);以0.1%甲酸水-0.1%甲酸乙腈为流动相进行梯度洗脱,体积流量为0.2 mL/min,柱温为40 ℃,进样量为2 μL。电喷雾质谱检测,正负离子模式下同时扫描获得高分辨质谱数据,通过文献和数据库结合质谱裂解规律的方式进行化合物鉴定。结果:在糖尿灵片的50%甲醇提取液中共鉴定得到100个化学成分,包括黄酮类、苯丙素类、三萜类、苯乙醇苷类、环烯醚萜苷类和酚类等化合物。结论:该研究首次采用UHPLC-Q-Exactive-Orbitrap-MS/MS法对糖尿灵片中的各类化学成分进行鉴定分析,为其药效物质基础和质量控制研究提供了依据。

[Abstract]
Objective: The purpose of the analysis was to determine the chemical composition of TangNiaoLing Tablets using UHPLC-Q-Exactive-Orbitrap-MS/MS. Methods: Separations by chromatography were carried out using a Waters ACQUITY HSS T3 column(100 mm × 2.1 mm, 1.8 μm). At a total flow rate of 0.2 mL/min, the mobile phase-which included an aqueous solution of 0.1% formic acid and acetonitrile mixed with 0.1% formic acid-was supplied. The injection volume was set at 2 μL and the column oven temperature was 40 °C. High-resolution mass spectrometric data were obtained by concurrently scanning the positive and negative ion ESI modes. And by inferring the mass spectrometric fragmentation pattern and comparing it with databases and references, the identification was accomplished. Results: The 50% methanol extract of TangNiaoLing pills included 100 different chemical elements, including triterpenes, flavonoids, phenylpropanoids, phenylethanoid glycosides, iridoid glycosides, and phenols, among others. Conclusion: The chemical contents of TangNiaoLing tablets were identified and analyzed using the UHPLC-Q-Exactive-Orbitrap-MS/MS method for the first time. This served as a foundation for future research into the tablets" effective components and quality control.
